Patienten mit PBC (Primary Biliary Cholangitis) werden eine Screening Visite bekommen, um zu bewerten, ob sie für die Studie geeignet sind. Das beinhaltet Bluttests, Elektrokardiogramme (EKGs), Atem- und Augentests.
Vier Wochen nach der ersten Visite kommen die Patienten erneut zur Bestätigung des Screenings.
Ab der Baseline Visite beginnen die Patienten mit der oralen Etrasimod Behandlung für 24 Wochen. Während der Behandlung wird es 8 klinische Visiten geben und 2 Wochen nach Ende der Studien-Behandlung wird es eine Nachfolgevisite (follow-up) geben.
Alle Visiten beinhalten EKGs und Bluttests. Bis zu 3 Visiten werden lange Visiten sein, um Blutproben zur Feststellung des Medikamentenspiegels zu entnehmen.
Bei jeder Klinikvisite werden Fragebögen über Juckreiz und die Lebensqualität ausgefüllt.
Die Patienten (und ihre Partner) müssen während der Studie eine Schwangerschaft verhüten.

Criteri per la partecipazione alla sperimentazione

- Männer und Frauen zwischen 18 und 80 Jahren (einschließlich) während des Screenings mit bestätigter PBC Diagnose auf Grundlage von zumindest 2 der folgenden 3 Kriterien:
o Mitochondriale Antikörper (AMA) Titer >1:40
o Alkalische Phosphatase (ALP) >1.5 x ULN (der normalen Obergrenze) für zumindest 6 Monate
o Leberbiopsie Befunde, die eine PBC bestätigen
- Unzureichende Reaktion auf Ursodeoxycholsäure (UDCA) nach 6 Monaten der UDCA Therapie, stabile Dosierung für die vorangegangenen 3 Monate vor dem Screening. Eine unzureichende Reaktion ist definiert als eine Erhöhung der Alkalischen Phosphatse im Blut um mehr als das 1,67-fache der Obergrenze des Normbereichs.
- Stabile Leberfunktions-Bluttest Ergebnisse, die innerhalb der vorgegeben Grenzwerte liegen. Die Tests werden im Zentrallabor durchgeführt.
Criteri di esclusione

- Chronische Lebererkrankungen, die keine PBC Ätiologie haben. PBC Patienten begleitet mit primärem Sjögren's Syndrom (pSS) sind geeignet für den Studieneinschluss. Keine signifikanten klinischen dekompensierten Leberschäden in der Krankengeschichte.
- Medizinische Gegebenheiten, die einen nicht-hepatischen ALP-Anstieg verursachen (z.B.Morbus Pagét)
- Klinisch signifikante Infektionen 6 Wochen vor dem Behandlungsbeginn oder Hepatitis C Infektionen in der Vergangenheit.
- Behandlung mit nicht lizenzierten Medikamenten oder mit Medikamenten, die eine Veränderung oder Unterdrückung des Immunsystems verursachen innerhalb von 30 Tagen vor Behandlungsbeginn. Behandlung mit Obeticholsäure (OCA) innerhalb von 30 Tagen vor Tag 1. Die Screening Periode wird zwischen 4-8 Wochen vor Tag 1 sein. Tag 1 ist der erste Tag, an dem ein Studienpatient das Studienmedikament bekommt.

Key Inclusion Criteria:
- Males or females aged 18 to 80 years (inclusive) at the time of screening, with
confirmed PBC diagnosis based upon at least 2 of 3 criteria:
- AMA titer >1:40 on immunofluorescence or M2 positive by enzyme-linked
immunosorbent assay (ELISA) or positive PBC-specific antinuclear antibodies
(anti-GP210 and/or anti-SP100)
- ALP >1.5 x ULN for at least 6 months
- Liver biopsy findings consistent with PBC
- Use of UDCA for at least 6 months prior to screening (stable dose for at least 3
months immediately prior to screening)
- Patients must have ALP >1.5 x ULN but
