Morbus Crohn (MC) ist eine chronische (langwierige) entzündliche Krankheit (einhergehend mit Reizungen oder Schwellungen), die alle Abschnitte des Verdauungstrakts betreffen kann. Zu den Symptomen zählen häufig Bauchschmerzen, Durchfall (der bei einer schweren Entzündung auch blutig sein kann), Fieber und Gewichtsverlust.
Das Ziel dieser Studie ist die Untersuchung der Wirksamkeit, Sicherheit und Verträglichkeit von Mongersen (GED-0301), um herauszufinden, ob dieses Prüfpräparat die Symptome von Morbus Crohn lindern kann oder nicht. Etwa 1064 Personen werden weltweit an dieser Studie teilnehmen.
In der Schweiz werden bis zu 24 Patienten teilnehmen.
In der Studie werden 2 Dosierungen von GED-0301 (160 mg und 40mg) im Vergleich zu Placebo (ein Scheinmedikament, das kein Prüfmedikament enthält) untersucht. Die Studienteilnehmer werden per Zufall einer von vier Gruppen zugeteilt. Die erste Gruppe erhält 160 mg GED-0301 über einen Zeitraum von 12 Wochen, gefolgt von einer Behandlung mit Placebo im Wechsel mit 160 mg GED-0301 alle 4 Wochen. Die zweite Gruppe erhält 160 mg GED-0301 über einen Zeitraum von 12 Wochen, gefolgt von einer Behandlung mit Placebo im Wechsel mit 40 mg GED-0301 alle 4 Wochen. Die dritte Gruppe erhält 160 mg GED-0301 über einen Zeitraum von 12 Wochen, gefolgt von einer Behandlung mit 40 mg GED-0301 über 40 Wochen. Die vierte Gruppe erhält Placebo für 52 Wochen. Die Studiendauer für Studienteilnehmer kann bis zu 60 Wochen betragen.

Criteri per la partecipazione alla sperimentazione

- Diagnose von MC mit einer Erkrankungsdauer von mindestens 3 Monaten vor dem Screeningtermin
- Vorliegen einer Ileitis, Ileokolitis oder Kolitis, diagnostiziert durch endoskopische, radiographische oder sonstige bildgebende Untersuchungen (z. B. Magnetresonanztomographie [MRT], Computertomographie [CT])
- Fehlschlagen oder Unverträglichkeit mindestens einer der folgenden Therapien: Aminosalicylate; Budesonid; systemische Kortikosteroide; Immunsuppressiva (d. h. Azathioprin [AZA], 6 Mercaptopurin [6 MP] oder Methotrexat [MTX]) oder Biologika für die MC Therapie

- Vorliegen einer der folgenden Diagnosen: Colitis ulcerosa (CU), nicht näher bestimmte Kolitis, ischämische Kolitis, mikroskopische Kolitis, Strahlenkolitis oder Divertikulitis assoziierte Kolitis
- Vorliegen lokaler MC Manifestationen wie Strikturen, Abszesse, Kurzdarmsyndrom; oder sonstige MC Komplikationen, aufgrund derer ein chirurgischer Eingriff angezeigt sein kann oder die Beurteilung der Wirksamkeit beeinflusst werden könnte
- Darmresektion innerhalb von 6 Monaten oder sonstiger Eingriff im Bauchraum innerhalb von 3 Monaten vor dem Screeningtermin

Inclusion Criteria:
Subjects must satisfy the following criteria to be enrolled in the study:
- Male or female = 18 years
- Active Crohn's disease (CD) disease as determined by the Crohn's Disease Activity
Index (CDAI) score and the Simple Endoscopic Score for Crohn's Disease (SES-CD)
- Must meet a determined average minimum number of daily stools or rating of abdominal
pain over a 7 day period
- Subject must have failed or experienced intolerance to at least one of the following:
budesonide; systemic corticosteroids; immunosuppressants (eg, azathiopurine,
6-mercaptopurine, or methotrexate); or biologics for the treatment of CD.
Exclusion Criteria:
The presence of any of the following will exclude a subject from enrollment:
- Diagnosis of ulcerative colitis (UC), indeterminate colitis, ischemic colitis,
microscopic colitis, radiation colitis or diverticular disease-associated colitis
- Local manifestations of Crohn's Disease (CD) such as symptomatic/severe strictures,
abscesses, short bowel syndrome; or other disease complications for which surgery
might be indicated or could confound the evaluation of efficacy
- Intestinal resection within 6 months or any intra-abdominal surgery within 3 months
prior to the Screening Visit
- Ileostomy or a colostomy
- Subject has a history of any clinically significant medical condition that, in the
investigator's opinion, would prevent the subject from participating in the study.
