Brief description of trial (Data source: BASEC)
Diese Studie ist eine Beobachtungsstudie, welche in der Schweiz durchgeführt wird. Benralizumab ist in der Schweiz unter dem Handelsnamen Fasenra® für die Behandlung von schwerem eosinophilem Asthma zugelassen. Die Zulassung von Fasenra® erfolgte erst Mitte 2018, daher sollen mit dieser Studie noch zusätzliche Daten gesammelt werden.
Es können alle Personen in der Schweiz teilnehmen, die an schwerem eosinophilem Asthma leiden und bei denen unabhängig von der Beobachtungsstudie entschieden wurde, eine Behandlung mit Benralizumab (Fasenra®) zu beginnen.
Das Hauptziel dieser Beobachtungsstudie ist die Beurteilung der von den Patienten berichteten Änderung der Asthma-Symptome nach Beginn der Behandlung mit Benralizumab (Fasenra®), um die Behandlung von schwerem Asthma zu optimieren. Es ändert sich nichts an Ihrer aktuellen Asthmatherapie, Sie werden im Studienverlauf nur genauer beobachtet und müssen verschiedene Fragebögen ausfüllen.

Benralizumab wird im Rahmen Ihrer Behandlung und unabhängig von der Studie als subkutane (unter die Haut) Injektion verabreicht, die ersten 3 Verabreichungen erfolgen alle 4 Wochen, danach alle 8 Wochen: Tag 1, Woche 4, Woche 8, Woche 16 Woche 24, Woche 32, Woche 40, Woche 48 und Woche 56.
Die Beobachtungstudie dauert für jeden Studienteilnehmer 56 Wochen und deckt somit die Dauer der ersten 9 Benralizumab Verabreichungen ab.
Die Teilnahme an der Studie erfordert folgende zusätzlichen Massnahmen:
- Zusätzliche Visiten Woche 1 und Woche 2: diese beiden Visiten können entweder als Besuch beim Arzt Woche 1 und Woche 2 oder via Telefon durchgeführt werden. Falls Sie sich für eine telefonische Visite entscheiden, werden Ihnen die Fragebögen nach Hause mitgegeben und Sie bringen die ausgefüllten Fragebögen bei Ihrem nächsten Besuch beim Prüfarzt zurück.
- Ausfüllen dreier kurzer Frageböogen mit insgesamt 7 Fragen zu Ihrer Erkrankung während der Visite Woche 0, 1, 2, 4, 8, 16 und 56.
- Ausfüllen eines kurzen elektronischen Tagebuchs zweimal täglich, morgens und abends vor der Einnahme Ihrer Asthma-Medikamente: dazu wird der Studienarzt mit Ihnen eine App auf Ihrem Handy herunterladen. Der Arzt wird Ihnen erklären, wie Sie das Tagebuch ausfüllen müssen.
- Messung des maximalen Ausatmungsflusses (Peak Flow) zweimal täglich, morgens und abends vor der Einnahme der Asthma-Medikamente. Sie erhalten zu diesem Zweck einen tragbaren Peak-Flow Meter. Der Studienarzt wird Ihnen die Handhabung des Peak-Flow Meters erklären.
- In der Woche 0, 8, 16 und 56 wird im Zentrum eine zusätzliche Spirometrie (Lungenfunktions-test) durchgeführt.
Criteria for participation in trial

1) Männliche oder weibliche Patienten, ab 18 Jahren und
mit einer vom Arzt bestätigten Diagnose von schwerem, unkontrolliertem Asthma
2) Asthma, das mindestens eine hochdosierte Inhalationstherapie (ICS + LABA) als Erhaltungsbehandlung erfordert.
3) Mindestens 2 Asthma-Krisen (Exazerbationen) in den letzten 12 Monaten
Exclusion criteria

1) Dokumentierte andere Lungenerkrankungen als Asthma, z.B. COPD, und nicht innerhalb der Zulassung liegen.
2) Aktuelle Teilnahme in einer anderen klinischen Studie, mit Ausnahme von:
- Patienten, die parallel in einem nationalen Asthma-Register dokumentiert sind.
- Patienten, die eine andere klinische Studie abgeschlossen haben

Inclusion Criteria:
Subject eligible for enrolment in the study and treatment for benralizumab according to the
specific Swiss label must meet all the following criteria:
1. Male or female patients older than 18 years with physician's confirmed diagnosis of
severe, uncontrolled asthma according to ATS/ERS guidelines.
2. Asthma requiring high-dose ICS plus LABA as maintenance treatment.
3. Minimum of 2 exacerbations in the last 12 months.
4. Documented peripheral blood eosinophil count = 300 cells/µL blood.
5. Provision of signed written informed consent form (ICF) indicating that they
understand the purpose of the study and procedures required for participation in the
study.
6. Patients must be able and willing to read and comprehend written instructions, and
comprehend and complete the questionnaires required by the protocol (ACQ-5, PGI-C and
PGI-S).
Exclusion Criteria:
Subjects meeting any of the following criteria will not be eligible to participate in the
study:
1. Documented lung diseases other than asthma, e.g. COPD, and not within reimbursed
label, e.g pregnancy or lactation.
2. Currently enrolled in an interventional clinical study in parallel, except:
- Patients being in parallel documented in a national asthma registry.
- Patients having completed any other clinical trial including those with biologic
treatment.
3. An acute or chronic condition that, in the investigator's opinion, would limit the
patients' ability to complete questionnaires or participate in this study or impact
the interpretations of results.
