Brief description of trial (Data source: BASEC)
Diese Studie umfasst eine Voruntersuchungsphase, eine Behandlungsphase, einen Besuchstermin bei Behandlungsende, einen Besuchstermin zur sicherheitsbezogenen Nachbeobachtung nach 30 Tagen und eine Langzeit-Nachbeobachtungsphase.
Der Patient kann DAY101 solange erhalten, wie sich die Krebserkrankung nicht verschlechtert, keine medizinisch unvertretbaren Nebenwirkungen auftreten und der zuständige Prüfarzt der Ansicht ist, dass der Gesundheitszustand eine Teilnahme an dieser Studie zulässt.
Die geschätzte Gesamtdauer dieser Studie wird für die einzelnen Teilnehmer voraussichtlich mindestens 2 Jahre betragen. Die Studie endet für alle Teilnehmer, wenn der zuletzt aufgenommene Patient seine 2-jährige Behandlung abgeschlossen hat.
Es gibt bestimmte Untersuchungen und Maßnahmen, die bei allen Patienten in dieser Studie erforderlich sind. Einige gehören zur Routineversorgung von Krebspatienten, werden jedoch vielleicht häufiger als üblich durchgeführt, da es sich hier um eine klinische Studie handelt. Andere dienen wiederum rein wissenschaftlichen Zwecken.
Damit man an dieser Studie teilnehmen können, muss das Vorliegen eines niedriggradig malignen Glioms bestätigt worden sein. Dies bedeutet, dass die Diagnose auf Zellen beruht, die dem Tumor entnommen wurden.

Criteria for participation in trial

Patienten im Alter von 6 Monaten bis 25 Jahren mit einem rezidivierenden oder progredienten LGG mit dokumentierter bekannter aktivierender BRAF-Alteration, die mithilfe molekularer Assays, wie sie routinemäßig in CLIA- oder anderen entsprechend zertifizierten Laboren durchgeführt werden, nachgewiesen wurde
Bestätigung der histopathologischen Diagnose eines LGG, d. h. entweder der ursprünglichen Diagnose oder eines Rezidivs/einer Progression
Die Patienten müssen mindestens eine systemische Therapielinie erhalten haben, und für den betreffenden Patienten muss der dokumentierte radiologische Nachweis einer Krankheitsprogression vorliegen.

Der Tumor des Patienten weist weitere, vorher bekannte aktivierende molekulare Veränderungen auf (z. B. Histon-Mutation, IDH1/2-Mutationen, FGFR-Mutationen oder -Fusionen, MYBL-Alterationen, NF1-Mutationen (somatisch oder in der Keimbahn))
Der Patient weist Symptome einer klinischen Progression ohne radiografisch nachweisbare rezidivierende oder progrediente Erkrankung auf.
Bekannte oder Verdachtsdiagnose einer Neurofibromatose Typ 1 (NF-1) mittels Gentest oder nach aktuellen Diagnosekriterien

Minimum age: 6 Months
Inclusion Criteria:
- Age 6 months to 25 years with:
1. Arms 1 & 2: a relapsed or progressive LGG with documented known activating BRAF
alteration
2. Arm 3: locally advanced or metastatic solid tumor with documented known or
expected to be activating RAF fusion
- Confirmation of histopathologic diagnosis of LGG and molecular diagnosis of activating
BRAF alteration
- Must have received at least one line of systemic therapy and have evidence of
radiographic progression
- Must have at least 1 measurable lesion as defined by RANO (Arms 1 & 2) or RECIST v1.1
(Arm 3) criteria
Exclusion Criteria:
- Patient's tumor has additional previously-known activating molecular alterations
- Patient has symptoms of clinical progression in the absence of radiographic
progression
- Known or suspected diagnosis of neurofibromatosis type 1 (NF-1)
- Other inclusion/exclusion criteria as stipulated by protocol may apply
