Brief description of trial (Data source: BASEC)
Lo studio prevede l'inclusione di pazienti sottoposti ad intervento chirurgico (LAR) negli ultimi 2 anni.
Sono previsti 2 periodi diversi di trattamento uno consecutivo all’altro. Il paziente assume Ondansetron (farmaco con principio attivo) due volte al giorno per 4 settimane e per le 4 settimane successive assumerà placebo (farmaco senza principio attivo) o viceversa. Tra i due trattamenti ci sarà una settimana di pausa.
Lo studio prevede anche la compilazione di questionari per valutare l'efficacia del trattamento e la qualità di vita del paziente.

Criteria for participation in trial

- età ≥ 18 anni
- pazienti in grado di seguire le procedure del protocollo
- donne potenzialmente fertili che non utilizzano un metodo contracettivo farmacologico devono acconsentire ad astenersi da rapporti sessuali durante il periodo di trattamento
- consenso informato firmato
- pazienti con tumore del retto che hanno subito un resezione anteriore bassa del retto (LAR) negli ultimi 2 anni e che sono trascorsi almeno 4 settimane dalla ricanalizzazione e 4 settimane dall'ultima chemio/radiotherapy
- presenza di anastomosi funzionale
- presenza di sintomi della sindrome di resezione anteriore bassa valutata tramite valutazione/punteggio LARS
Exclusion criteria

- conoscenza di ipersensitività o allergia ad ondansetron
- precedente terapia antiemetica con ondansetron nelle 4 settimane prima dell'inizio del trattamento in studio
- chemio/radioterapia antitumorale completata nelle 4 settimane prima dell'inizio del trattamento in studio
- sindrome congenita dell'allungamento del Q-T
- ipopotassiemia e ipomagnesiemia non corretta, solo in pazienti con fattori di rischio o sintomi clinici significativi
- donne incinta, che allattano o che potrebbero restare incinta durante il trattamento
- malattie concomitanti significative o complicazioni anastomotiche che potrebbero impedire al paziente la partecipazione allo studio
- incapacità di seguire le procedure dello studio (problemi di lingua, condizioni psicologiche, demenza, ecc.)

Inclusion Criteria:
- Age = 18 years
- Patients willing and able to comply with the study procedures
- Female patients of childbearing potential must agree to use a reliable method of
contraception
- Written informed consent Patients who have undergone low anterior resection (LAR) for
rectal cancer in the last 2 years provided that at least 4 weeks have elapsed since
recanalization and 4 weeks have elapsed since any chemo- and/or radiotherapy
- Presence of functioning anastomosis
- Presence of significant LARS symptoms assessed by LARS Score
Exclusion Criteria:
- Known hypersensitivity or allergy to Ondansetron
- Previous Ondansetron antiemetic therapy completed less than 4 weeks before study entry
- Antitumor chemotherapy or radiotherapy completed less than 4 weeks before study entry
- Congenital long Q-T syndrome
- Ongoing treatment with drugs causing prolongation of the Q-T interval
- Uncorrected hypokalemia or hypomagnesemia
- Women who are pregnant or breast feeding or are willing to become pregnant during the
study
- Clinically significant concomitant disease states or anastomotic complications which
could impair the ability of the patient to participate in the trial
- Inability to follow the procedures of the study, e.g. due to language problems,
psychological disorders, dementia, etc.
