Brief description of trial (Data source: BASEC)
Ziel dieser Studie ist es zu untersuchen, ob durch die Einnahme des Studienmedikaments Macimorelin das Antidiuretische Hormon (ADH) stimuliert werden kann. Das ADH ist das wichtigste Hormon für die Regulierung des Wasser- und Salzhaushaltes im Körper. Bestimmte Krankheiten des Wasser- und Salzhaushaltes (Diabetes insipidus und Primäre Polydipsie) gehen mit einer Störung der Ausschüttung des ADH‘s einher. Um diese Krankheiten zu diagnostizieren ist aktuell ein mehrstündiger aufwändiger Test notwendig, bei welchem eine 3% NaCl-Infusion erfolgt und regelmässige Blutkontrollen notwendig sind um im Blut die Stimulation auf das ADH zu messen. Da das ADH schwierig messbar ist, wird stattdessen Copeptin, ein Nebenprodukt des Vorläuferhormons des ADH, im Blut gemessen.
Macimorelin ist als Trinklösung verfügbar und wird bereits in der Diagnostik des Wachstumshormonmangels eingesetzt, weil es die Ausschüttung von Wachstumshormon stimuliert.
Wir wollen mit dieser Studie nun untersuchen, ob Macimorelin nicht nur Wachstumshormon sondern auch ADH/Copeptin und andere Hormone stimuliert. Sollten sich diese Hormone durch Macimorelin stimulieren lassen, könnte in Zukunft die Diagnostik bei Störungen des Wasser-/Salzhaushaltes deutlich vereinfacht werden, durch diesen neuen Test.

Untersucht wird ein diagnostischer Test mit der Trinklösung Macimorelin. Macimorelin wird zur Diagnosestellung eines Wachstumshormonmangels verwendet. In dieser Studie untersuchen wir, ob Macimorelin auch auf Copeptin und andere Hormone stimulierend wirkt.

Criteria for participation in trial

- gesunde Personen
- 18-60 Jahre alt
- keine Einnahme von Medikamenten (exkl. Verhütungs-Pille)
Exclusion criteria

- BMI < 18.5kg/m2 oder > 40kg/m2
- Eine Trinkmenge > 3l/Tag und eine gleichzeitig eine erhöhte Urinausscheidung von > 50ml/kg Körpergewicht
- ein verlängertes QT-Intervall in der elektronischen Herzkurve

- No medication except hormonal contraception
Exclusion Criteria:
- Body Mass Index (BMI) > 40kg/m2 or BMI < 18.5 kg/m2
- participation in a trial with investigational drugs within 30 days
- vigorous physical exercise within 24 hours before the study participation
- Alcohol intake within 24 hours before study participation
- pregnancy and breastfeeding
- Evidence of disordered drinking habits and diuresis defined as polyuria >50ml/kg body
weight/24h and polydipsia >3l /24h
- a prolonged QT interval (QTc >500 ms) or concomitant treatment with drugs that prolong
the QT/QTc.
- Intention to become pregnant during the course of the study
- Known allergy towards Macimorelin
