Das Hauptziel dieser Studie ist es zu untersuchen, wie lange Patienten mit Glioblastom (ein bösartiger Hirntumor) leben, die Temozolomid plus Strahlentherapie erhalten, im Vergleich zu Patienten, die mit Nivolumab in Kombination mit Temozolomid plus Strahlentherapie Strahlentherapie behandelt werden

- Experimenteller Behandlungsarm (Medikament): Nivolumab intravenös (als Infusion) 16 Wochen lang im 2-wöchigen Abstand, ab Woche 17 weiterhin im 4-wöchigen Abstand.
Temozolomid 75 mg/m2 täglich als Tablette während der Strahlentherapie, gefolgt von einer 4-wöchigen Behandlungspause. Im Anschluss 150 mg/m2 als Tablette an 5 Tagen die Woche (Tag 1-5) für 1 Woche dann 200 mg/m2 an 5 Tagen die Woche (Tage 1-5) bis einschließlich Woche 6, soweit verträglich.
Strahlentherapie (2 GY) an 5 Tagen die Woche für 6 Wochen.
- Vergleichsarm (Plazebo):

Criteria for participation in trial

- Männer und Frauen im Alter von mindestens 18 Jahren
- Neu-diagnostiziertes Glioblastom (Hirntumor)
- Der Tumor ist vom methylierten, partiell-methylierten, oder nicht bestimmbaren MGMT-Typ (dafür wird eine Laboruntersuchung des Tumormaterials gemacht)
- Vollständige Erholung von der Tumor-Operation
- Leistungsstatus nach Karnofsky von mind. 70 (d.h. die Person ist in der Lage sich selbst zu versorgen)
Exclusion criteria

- Vorherige Therapie des Glioblastoms (mit Ausnahme einer operativen Entfernung)
- Eine Tumorerkrankung ausserhalb des Gehirns
- Wiederkehrendes oder sekundäres Glioblastom
- Bekannte aktive Autoimmunerkrankung (oder Verdacht auf eine solche Erkrankung)
- Tumorentfernung durch Biopsie, bei der nur bis zu 20% der Tumormasse entfernt wurde

Inclusion Criteria:
- Males and Females, age = 18 years old
- Newly diagnosed brain cancer or tumor called glioblastoma or GBM
- Karnofsky performance status of = 70 (able to take care of self)
- Substantial recovery from surgery resection
- Tumor test result shows MGMT methylated or indeterminate tumor subtype
Exclusion Criteria:
- Biopsy-only of GBM with less than 20% of tumor removed
- Prior treatment for GBM (other than surgical resection)
- Any known tumor outside of the brain
- Recurrent or secondary GBM
- Active known or suspected autoimmune disease
