Brief description of trial (Data source: BASEC)
Das Ziel dieser Studie ist, festzustellen, ob die Kombination von Nivolumab und Ipilimumab eine bessere Wirksamkeit aufweist als Nivolumab bei Patienten mit Melanom im Stadium IIIb/c/d oder IV nach vollständiger Resektion.
Die Studie besteht aus drei Phasen: Voruntersuchungs-, Behandlungs- und Nachbeobachtungsphase.
In der Voruntersuchungsphase wird die Eignung der Patienten für Studienteilnahme überprüft. Diese Phase dauert ca. 1 Monat.
In der Behandlungsphase werden die Studienteilnehmer nach dem Zufallsprinzip einer Behandlungsgruppe (Arm A, B oder C) zugewiesen und entsprechend Nivolumab in Kombination mit Ipilimumab , Nivolumab- Monotherapie oder Ipilimumab- Monotherapie erhalten. Diese Studie ist verblindet, das bedeutet, dass die Studienteilnehmer nicht erfahren, welcher Behandlung sie zugeteilt sind.
Jeder Teilnehmer an dieser Studie wird die verblindete Behandlung für bis zu 49 Wochen (ca. 1 Jahr) erhalten.
Die Nachbeobachtungsphase beginnt nach dem Absetzen oder Beendigung der Behandlung mit der Studienmedikation. Diese Nachbeobachtungsphase kann bis zu 5 Jahre nach der ersten Medikamentengabe dauern. Während diese Phase wird der Gesundheitszustand der Studienteilnehmer weiter verfolgt.

Criteria for participation in trial

• Eine vollständige Resektion des Melanoms im Stadium IIIb/c/d oder IV muss innerhalb von 12 Wochen vor der Randomisierung durchgeführt werden.
• Die Studienteilnehmer müssen unbeschränkt körperlich aktiv sein oder mindestens in der Lage sein zu Laufen und leichte Hausarbeit sowie Büroarbeit zu erledigen.
• Die Studienteilnehmer dürfen für ihr Melanom keine Antikrebstherapie (mit Ausnahme von operativer Entfernung der Melanoms Läsionen und/oder adjuvanter Bestrahlung nach Resektion der ZNS-Metastasen)
erhalten haben.
Exclusion criteria

• Uvealmelanom in der Vorgeschichte
• Gewicht ≤ 40 kg
• Studienteilnehmer mit einer aktiven, bekannten oder vermuteten Autoimmunerkrankung
• Vorherige Therapie eines Melanoms mit Interferon (wenn sie < 6 Monate vor der Randomisierung abgeschlossen wurde), Anti-PD-1-, Anti-PD-L1-, Anti-PD-L2-, Anti-CD137- oder Anti-CTLA-4-Antikörpern oder andere Antikörper oder Medikamente, die speziell die TZell- Costimulation oder Checkpointpfade angreifen

Inclusion Criteria:
- Completely surgically resected stage IIIb/c/d or stage IV melanoma within 12 weeks of
participation in study.
- Must have full activity or, if limited, must be able to walk and carry out activities
such as light house work or office work
- No prior anti-cancer treatment for melanoma (except surgery for the melanoma lesion(s)
and/or except for adjuvant radiation therapy (RT) after neurosurgical resection for
central nervous system (CNS) lesions)
Exclusion Criteria:
- History of uveal melanoma
- Patients with active, known or suspected autoimmune disease
- Prior treatment with interferon (if complete < 6 months prior to participation in
study), anti-PD-1, anti-PD-L1, anti-PD-L2, anti-CD137, or anti-CTLA-4 antibody, or any
other antibody or drug specifically targeting T-cell co-stimulation or checkpoint
pathways
