Morbus Crohn (MC) ist eine lang anhaltende, entzündliche Erkrankung von verschiedenen Darmabschnitten. MC kann Müdigkeit, Durchfall (mit oder ohne Blut im Stuhl), Bauchschmerzen, Gewichtsabnahme und Fieber verursachen.
Risankizumab ist ein Studienmedikament, das zur Behandlung entzündlicher Erkrankungen entwickelt wurde. An dieser Studie können Patienten mit mittelschwerem bis schwerem MC teilnehmen, die auf eine vorhergehende Therapie mit einem Biologika nicht angesprochen haben (zum Beispiel Humira®, Remicade®, Cimzia®, Tysabri®, Entyvio®).
In dieser Studie wird die Wirksamkeit und Sicherheit von Risankizumab im Vergleich zu Placebo (Medikament ohne Wirkstoff) als Induktionstherapie untersucht. Die Induktionstherapie dient dazu, Anzeichen und Symptome einer Erkrankung unter Kontrolle zu bringen. Patienten, welche auf die Behandlung ansprechen, können an einer Nachfolgestudie teilnehmen.

Etwa 579 Patienten mit MC werden weltweit an dieser Studie teilnehmen.
Die Studie umfasst eine oder zwei Induktionsphasen. In den ersten 12 Wochen (Induktionsphase 1) werden die Patienten zufällig in eine von drei verschiedenen Behandlungsgruppen zugewiesen: 2 Gruppen mit Risankizumab (mit 2 verschiedene Dosierungen) und 1 Gruppe mit Placebo (Medikament ohne Wirkstoff). Die Studie ist verblindet, das heisst, dass weder der Patient noch der Arzt wissen, welche Behandlung der Patient erhält.
Wenn an Woche 12 die Behandlung nicht gewirkt hat (kein klinisches Ansprechen), werden die Patienten neuen Behandlungsgruppen zugewiesen (Induktionsphase 2, dauert ebenfalls 12 Wochen): Patienten, welche bereits Risankizumab erhalten haben, werden in eine von drei verschiedenen Gruppen mit Risankizumab eingeteilt (drei Dosierungen). Patienten, die im ersten Teil Placebo bekommen haben, erhalten nun Risankizumab (eine Dosierung).
Patienten, die in Woche 12 (Induktionsphase 1) oder Woche 24 (Induktionsphase 2) auf die Behandlung angesprochen haben, können an einer Nachfolgestudie teilnehmen.

1. Männer oder Frauen, die zwischen 18 und 80 Jahre alt sind mit einer Morbus Crohn Diagnose mindestens 3 Monate vor der ersten Einnahme des Studienmedikamentes.
2. Bestätigte Diagnose von mittelschwerem bis schwerem aktivem MC, beurteilt durch die Stuhlfrequenz, der Intensität der Bauchschmerzen und der Auswertung einer einfachen Endoskopie für MC.
3. Unverträglichkeit gegenüber oder unzureichendes Ansprechen auf biologische Therapien für MC.

1. Patienten mit aktuell diagnostizierter Colitis ulcerosa oder unbestimmter Colitis.
2. Patienten mit unregelmässiger Medikamenteneinnahme zur Behandlung des MC.
3. Erhalt von MC anerkannten biologischen Therapien innerhalb von 8 Wochen vor der ersten Einnahme des Studienmedikamentes.
4. Patienten, welche bereits eine Behandlung mit einem p40-Inhibitor (z.B. Stelara®) oder p19-Inhibitor (z.B. Risankizumab) erhalten haben.
5. Patienten mit aktuell bekannten Komplikationen des MC (z. B. Strikturen, Stenosen, Kurzdarmsyndrom usw.).

Inclusion Criteria:
- Male or female aged >=18 to <= 80 years, or minimum age of adult consent according to
local regulations, at the Baseline Visit. Where locally permissible, participants 16
to < 18 years of age who meet the definition of Tanner stage 5 for development at the
Baseline Visit.
- Confirmed diagnosis of CD for at least 3 months prior to Baseline.
- Crohn's disease activity index (CDAI) score 220 - 450 at Baseline.
- Confirmed diagnosis of moderate to severe CD as assessed by stool frequency (SF),
abdominal pain (AP) score, and Simple Endoscopic Score for Crohn's Disease (SES-CD).
- Demonstrated intolerance or inadequate response to biologic therapy for CD.
- If female, participant must meet the contraception recommendations.
Exclusion Criteria:
- Participant with a current diagnosis of ulcerative colitis or indeterminate colitis.
- Participants with unstable doses of concomitant Crohn's disease therapy.
- Receipt of Crohn's disease approved biologic agents (infliximab, adalimumab,
certolizumab, vedolizumab, natalizumab within 8 weeks prior to Baseline or ustekinumab
within 12 weeks prior to Baseline), or any investigational biologic or other agent or
procedure within minimally 35 days or 5 half-lives prior to Baseline, whichever is
longer.
- Prior exposure to p19 inhibitors (e.g., risankizumab).
- Complications of Crohn's disease.
- Having an ostomy or ileoanal pouch.
- Known active Coronavirus Disease 2019 (COVID-19) infection.
